Sue has a strong commitment to research focusing on interventions for disease control in the Global South, with on-going projects in Kenya, Morocco, Mozambique, Tanzania, Uganda, Nigeria, Zambia and Zanzibar. Her research has resulted in over 195 published works including 153 original research papers, 31 peer refereed reviews and 11 book chapters arising from >£30 million in research support.

Her publications have been cited over 6500 times and her h-index is 44.  Sue has supervised 49 PhD and 4 MSc research students from Europe, the Americas, Africa and Asia, working on all aspects of Zoonotic disease management. Sue has a strong commitment to career development of young scientists. She was awarded the Chancellor’s Medal in 2011 by HRH The Princess Royal, Chancellor of The University of Edinburgh for her work on development of research-led distance learning MScs.

正在开展的课题:

1.   CD8+ T细胞亚群与自身免疫性疾病与抗肿瘤免疫:  CD8+调节性T细胞在免疫调节中起着关键的作用。我们最新的研究表明,CD8+调节性T细胞的功能受到抑制性受体CD94/NKG2A的严格控制。通过对其配体Qa-1进行点突变阻断CD94/NKG2A的信号,能释放出CD8调节T细胞的活性,可用于自身免疫性脑脊髓炎(EAE)的治疗(Lu et al. PNAS 2008)。最新的研究表明,多发性硬化症治疗药物GA(Glatiramer acetate ) 能在病人体内诱导调节性CD8+ T细胞。我们最近发现GA在小鼠中也能诱导这类CD8+ T细胞。本实验室在小鼠EAE和IBD疾病模型中进一步研究GA诱导的CD8+调节性T细胞的特性和作用,并在在小鼠中建立一个基于CD8+调节性T细胞诱导的自身免疫病治疗策略(Yao Y et al. Eur. J. Immunol. 2013)。最近,我们正在研究一类CD8+ T细胞亚群在免疫耐受、抗肿瘤和抗病毒免疫反应中的作用。

2.胸腺T细胞的发育分化:利用生物信息学技术,克隆了一系列胸腺T细胞特异性表达的基因,进一步研究其在T细胞发育和功能调控中的作用,发现了一个与T细胞发育相关的关键调控因子Tespa1(Wang D et al. Nature Immunology 2012)。我们正在此基础上,深入研究Tespa1调控TCR信号传导和胸腺T细胞发育的机制。实验室也在针对另外几个重要的分子进行研究,以期揭示这些分子在免疫系统发育分化, 免疫反应调节和自身免疫性疾病中的功能,并开展分子机制的研究。

3.T细胞分化的信号传导调控:本实验室在前期的研究中发现MAPK4家族激酶MINK1可以通过感受细胞的氧化还原状态,调节T细胞的分化。在此基础上,我们正在研究T细胞分化过程中此类激酶的调控作用,以及微环境信号如何和TCR及细胞因子信号整合的分子机制。

郭博士于1992年在中国天津南开大学获得微生物学学士学位,1996年在北京中国科学院微生物所获得生物化学硕士学位,2002年在美国德克萨斯大学休斯敦M.D.Anderson癌症中心获得分子遗传博士学位。2003-2010年在加州大学洛杉矶分校吴虹实验室从事博士后研究。2010年加入清华大学做tenure-track副教授, 2017年加入浙江大学海宁国际校区做tenured副教授。郭博士专注于造血干细胞和肿瘤耐药的研究。郭伟实验室精于人类和小鼠遗传学,分子生物学,细胞生物学,生物医学,及基因组学的方法。郭博士在造血干细胞的损伤反应、白血病发病、肺癌干细胞、大肠癌转移以及肿瘤耐药调控等方面有突破性建树。他建立了第一个由PTEN缺失导致T细胞急性白血病的小鼠模型,揭示了该种新亚型的存在,多因子突变导致白血病干细胞形成的过程,以及mTOR抑制剂的双重抗白血病机制。而且分离了临床肿瘤中的肺癌干细胞,揭示NOTCH1调控其自我更新和铂类耐药的机制。郭博士在Nature、Cell Stem Cell、PNAS、Blood、Cancer Research、Molecular Cell Biology等国际权威期刊上发表了十余篇重要的论文。由于郭博士的杰出研究工作,他被邀请到世界著名的研究机构和会议作学术演讲,并且受邀做Stem cells、oncotarget等著名期刊的论文评审人以及国家自然基金委、欧洲捷克共和国等的基金评审人。
